Time, and the associated increase in entropy, is the greatest villain: inexorable, patient, untouchable because woven in to the very fabric of existence (spacetime). It eventually destroys everything.
Who is the worthy adversary? Intelligent, self-repairing life, able to fight back indefinitely against changing conditions.
Personify the struggle.
